LL-37 Australia: what is the human cathelicidin

It is the only member of the cathelicidin family of antimicrobial peptides made by humans. It is released from the tail of a larger precursor, hCAP18, coded by the CAMP gene, and takes its name from its length, 37 amino acids, and its first two residues, both leucine. The gene and the processing of the precursor were described in the European Journal of Biochemistry in 1996. The peptide carries a strong positive charge at neutral pH and has no cysteines. Neutrophils make it, as do the cells lining skin, gut and airways.

LL-37 peptide: how it works in research

In water the peptide is largely unfolded. On meeting the negatively charged membranes of bacteria it folds into a helix with a water-loving face and a fat-loving face, and that shape lets it disrupt the membrane directly. Beyond that physical action it binds lipopolysaccharide, the outer coat of many bacteria, and acts as a signal to immune cells. A 2000 study in the Journal of Experimental Medicine showed it draws human neutrophils, monocytes and T cells through the receptor then called FPRL1, now FPR2. Laboratories use it to study membrane disruption, biofilms, lipopolysaccharide neutralisation and immune cell movement.

LL-37 research: what the cathelicidin studies show

After the 1996 gene paper, a 2001 study in Blood identified proteinase 3 as the enzyme that cuts hCAP18 to release the active peptide outside neutrophils. The 2000 chemotaxis paper used human blood cells in culture. A 2006 review in Biochimica et Biophysica Acta summarised its structure, membrane interactions and wider biological roles.
